● 可以将逆转录病毒中的目的基因高效转入造血干细胞、T淋巴细胞等哺乳动物来源的细胞。 ● 避免感染过程中因使用polybrene造成的细胞毒作用。 ● 同样适用于感染常规的细胞系。 ● RetroNectin是重组人纤维连接蛋白片段,包括细胞结合域,肝磷脂结合域和CS1位点三个功能区域。它由574个氨基酸组成,分子量63 kDa。RetroNectin能够大大的提高逆转录病毒感染哺乳动物细胞的感染效率,其作用原理是:细胞表面VLA-4及VLA-5可分别于RetroNectin上的CS-1位点及细胞结合域结合,逆转录病毒载体可以与肝磷脂结合域结合。在经RetroNectin包被的培养容器表面,细胞与病毒载体以高浓度共存,从而提高基因转染效率30%-70%。 ● 另外RetroNectin还可以刺激淋巴细胞增殖,尤其是胸腹水以及肿瘤组织分离的TIL细胞,利用RetroNectin技术均可使其获得上千倍的增殖,已有部分客户将RetroNectin技术扩增TIL的方法进行专利申请。 |
